Quantification is possible by a modified van Urk reaction with vanillin reagent in sulphuric acid solution:
dry plate at ambient temperature in the dark in an air stream
mark spots at 254 nm under UV and scrape off marked zones
in separate flasks mix scraped material with 5% sulphuric acid in methanol, shake 5 min
add freshly prepared reagent solution (0.125 g vanillin in 100 ml 70% aqueous sulphuric acid), cooling and shaking constantly
let stand in the dark, shake occasionally, then filter
measure adsorption at 545 nm in 2 cm glass cells
